Heterologous und homologous overexpression of proteins
The production of the necessary high amounts of proteins for spectroscopy is performed in the bacteria Escherichia coli, Rhodobacter sphaeroides and Halobacterium salinarum. The respective genes are introduced into the bacterium with the help of vectors. The bacterium is then cultivated on media and triggered for overproduction of the protein. After the cells are harvested and lysed, the protein is purified by ultracentrifugation and column chromatography.
